Shear Stimulation of Adult Mesenchymal Stem Cells Towards Adipogenic Lineage

Abstract

Plated bioreactor systems are commonly used to shear stimulate adult human mesenchymal stem cells (hMSCs). æPreceding shear stimulation work has failed to investigate the differentiation of hMSCs in an in vivo-like environment. æWith a microfluidic approach, the ability of hMSCs to differentiate in a suspension-based tubing system was studied. æhMSCs of multiple passages and various lengths of tubing were analyzed using flow cytometry, qRTPCR, and immunohistochemical staining. æThe differentiation of the hMSCs in the suspension-based tubing system occurred without the use of a bioreactor system or specialized differentiation medium. æThis study demonstrates the capabilities of hMSCs to differentiate towards the adipogenic lineage through a shear stimulation method of reduced costs and time.
