Pathological states that are characterized by qualitative temporal changes of physiological variables have been coined Dynamics Diseases. Here I define diseases more generally as abnormal spatio-temporal patterns arising from complex physiological networks. Epileptic seizures are discussed as a special case of chronic brain disorders for which spatio-temporal clinical data are available. I present approaches to the mechanistic modeling of the transition into and out of seizure dynamics. Using imaging-inferred connectivities, computational models can be adapted to a patient’s individual network features. They then allow efficient testing of patient-specific hypotheses about pathological mechanisms. Specifically, there is now interest in novel non-drug treatments via bespoke interaction with the disease network.
